![]() ![]() In this perspective, we focus on the analysis of multiple DNA adducts within a sample with the emphasis on untargeted analysis. However, for the simultaneous detection, identification and quantification of both known and unknown DNA adducts, mass spectrometry (MS) is deemed to be the most promising technique. Over the years, a range of techniques and methods have been developed to study the formation of endo- and exogenous DNA adducts. The study of DNA adduct types and levels in human tissue therefore offers an interesting tool in several fields of research, including toxicology and cancer epidemiology. En outre, plusieurs tests de cytotoxicité et de mutagénicité ont été réalisés : le test MTS, le test CellToxTM Green et le test d’Ames. Ces résultats prouvent, pour la première fois, l’interaction spontanée et directe entre l’ADN et nos 7 oxy-PAH. Des adduits ont été détectés en post-marquage au phosphore 32 puis ont été identifiés et caractérisés en spectrométrie de masse. Une liste de 7 oxy-HAP, considérés comme prioritaires, a été sélectionnée sur des critères de présence dans l’environnement et de possible toxicité pour l’homme.Afin d’évaluer le potentiel mutagène et génotoxique de ces oxy-HAP prioritaires, de la 2’-désoxyguanosine (2’-dG), de l’ADN du thymus de veau et des cellules épithéliales bronchiques humaines immortalisées (BEAS-2B) ont été exposées à ces molécules. Ce travail s’est donc axé sur les oxy-HAP car, bien que ces composés soient omniprésents dans l'environnement, ils restent encore largement sous-étudiés en termes de toxicité. Des études de toxicité ont démontré que les HAP oxygénés (oxy-HAP) et les HAP nitrés (nitro-HAP) étaient responsables d'une grande partie de la toxicité totale des HAP. Its success in fully reaching this potential will depend on the application of validated biomarkers, with adherence to sound epidemiologic and ethical principles. Finally, molecular epidemiology has enormous potential in cancer prevention through the early identification of 'at risk' populations and the rapid assessment of intervention efficacy. Another emerging area of molecular epidemiology concerns the role of nutrition and specific dietary factors (including studies on antioxidants, energy metabolism, insulin and various growth factors) and the modulating effect of genetic polymorphisms. Recent advances in genomics, microassay technologies and informatics hold promise for rapid identification of polymorphic variants or changes in expression of genes influencing both response and susceptibility to carcinogens. There is also increasing recognition that individual risk from carcinogen exposure varies as a function of both inherited and acquired factors. Sensitive, validated biomarkers are needed to detect these mechanisms in small numbers of cells, tissues or fluids. The mechanisms include aberrant DNA methylation, oxidative damage, effects on metabolism of nitrogen oxide and nitrites, activation of receptors and transcription factors, cyclins and other cell cycle proteins. Although much valuable progress continues to be made in the study of carcinogens that cause direct DNA damage and are mutagenic, exogenous and endogenous carcinogens can also act by altering gene expression, cell proliferation and differentiation. However, a review of major findings to date reveals several urgent research needs to keep pace with the rapid evolution in knowledge of mechanisms in carcinogenesis. There have been major advances, notably in the understanding of environment-susceptibility interactions in human cancer. Here, we review progress during the past 17 years in validating and applying this approach to cancer prevention. In 1982 we proposed the concept and a framework for implementing molecular cancer epidemiology.
